Leveraging Genetics to Improve Personalized Prostate Cancer Risk Assessment

To improve intermediate-high-grade prostate cancer risk prediction, we will assess the extent to which incorporating genetically adjusted PSA (PSAG) and a polygenic risk score for aggressive disease (PRSAgg) into the Kaiser Permanente Prostate Cancer Risk Calculator improves prediction of higher-grade prostate cancer at biopsy. This work will include risk modeling in a cohort of Kaiser Permanente members who have undergone prostate biopsy and have existing genetic data in the Kaiser Permanente Research Bank. We also aim to improve prediction of prostate cancer active surveillance outcomes by evaluating the extent to which PSAG and PRSAgg improve the validated Canary PASS Active Surveillance Risk Calculator for prediction of prostate cancer progression on follow-up biopsies in men on active surveillance.
